Oxygen production through photosynthesis is likely one of the most significant events in the transformation of the planet by living organisms throughout geological time (Figure 1). A team from the LCB, in collaboration with the BIAM/LGBP and Clément Car, investigated the role of oxygen in the evolution of photosymbioses across macro- and microevolutionary scales (1), which remains largely poorly understood (2).

Insights into the biotic and abiotic environments that accompanied ancestral processes shed light on the biology of contemporary or establishing photosymbioses. This knowledge can also inform us about the evolutionary forces (natural selection, genetic drift, mutation, and gene flow) shaping photosymbioses and provide clues about ancestral events, which remain inherently impossible to observe directly. An experimental approach was used in a previous paper to suggest that oxygen provision preceded carbon supply as a selective force in the evolution of photosymbioses (3).

In this review (1), the authors first outline the benefits hosts can derive from the O₂ produced by their photosymbionts. They then examine the wide array of mechanisms that mitigate the oxidative stress associated with this O₂ production. Finally, the review discusses how present-day hypoxic zones represent relevant ecosystems for studying the mechanisms underlying ancestral and contemporary photosymbioses, as well as how photosynthetic O₂ can influence their evolution, particularly through niche construction processes.

As recent global change continues to expand the extent of hypoxic aquatic environments and their massive impact on ecosystems, studying how photosymbioses establish and evolve within them becomes critical to better anticipating the transformation of these environments.

This review echoes a popular science article (4) published in The Conversation and featured in Science & Vie (5).

With its intriguing social life style, Myxococcus xanthus is one of the most well-known examples of predatory bacteria. These bacteria form multicellular swarms that – like microscopic wolf packs – collectively hunt other microbes, form intricate wave patterns while feeding, and transform into elaborate fruiting bodies when they starve. In fruiting bodies, the rod-shaped vegetative cells differentiate into spherical spores that are resistant to environmental stresses. Under favourable environmental conditions a single fruiting body can regenerate a full colony of M. xanthus and thus restart the life cycle. It is essential for the survival of a swarm that germination only happens if sufficient nutrients are available. Yet, fruiting bodies are non-motile and consist mostly of dormant spores. This poses the question of how changes in environmental conditions are monitored to allow fruiting bodies to germinate under the right conditions. We are combining different imaging techniques and transcriptomics to study the germination behaviour of transplanted M. xanthus fruiting bodies at the collective and single cell scales. Our preliminary data from transplantation experiments at the collective scale suggest that germination can be induced by the presence of prey in the vicinity of fruiting bodies, while a direct contact to prey is not necessary. On the contrary, E. coli prey colonies can induce germination at a distance, which is then followed by an apparently directed expansion of the newly formed swarm towards the prey colony.

A study published in Nature Communications reveals an unexpected protein architecture that is crucial for bacterial respiration. In Bacillus subtilis, type II NADH dehydrogenase, long considered to be an enzyme simply associated with the membrane, is in fact organized around a tubular scaffold formed by the YjlC protein. This hydrophobic tube, filled with lipids and quinones, acts as an extension of the membrane and allows the enzyme to efficiently access the molecules that mediate electron transfer.

This work uncovers a novel supramolecular assembly that can stack into fibres and is essential for respiratory activity. It broadens our understanding of the organization of bacterial respiratory chains and reveals the role of specialized protein modules in managing electron flow.

The study was led by Axel Magalon’s team at the LCB, with contributions from Pierre Garcia from the Py team, researchers in Grenoble, and the technological platforms of the UAR.
To survive nutrient deprivation, sporulation is one of the strategies employed by some Gram-positive bacteria. This cellular differentiation leads to the formation of an endospore resistant to a range of harsh conditions, such as heat, UV radiation, desiccation and chemical agents. This confers extensive resilience on sporulating bacteria, some of which are pathogenic and still represent a public health concern. Decades of research into its development in the non-pathogenic model organism Bacillus subtilis have revealed numerous transcriptional and post-translational regulatory mechanisms.

However, several studies suggest that post-transcriptional regulation also plays an important role in this process. Although some RNA-binding proteins (RBPs) and non-coding RNAs have been studied, their overall contribution remains poorly understood. To address this, we adapted the high-throughput approach of OOPS (Orthogonal Organic Phase Separation) for the first time in B. subtilis to specifically purify the RNA-binding proteome (RBPome). This strategy enabled us to track the dynamics of the RBPome during sporulation and reveal major rearrangements. Consequently, we have identified novel candidate RNA-mediated post-transcriptional regulators of sporulation. Selected proteins are currently being further characterized to elucidate their functions and RNA targets

Iron-sulfur clusters (Fe-S) are critical cofactors required for the activity of many proteins, including components of the oxidative phosphorylation chain, metabolic enzymes, transcription factors, proteins involved in biogenesis of ribosomes and replication/repair of DNA. The Fe-S biogenesis pathway, termed ISC, which functions in bacteria and mitochondria, is complex, involving many dedicated proteins. Among them is a specialized chaperone system consisting of a J-domain protein (JDP) co-chaperone and its Hsp70 partner (termed HscB and HscA in Escherichia coli, respectively). Over the years detailed biochemical studies revealed that HscB/HscA chaperones interact with a scaffold protein, IscU, on which FeS are synthesized, using a binding cycle typical for all JDP/Hsp70 systems. Yet, despite this detailed analysis, little is known about how IscU binding translates into Fe-S biogenesis in vivo. In this seminar, I will present our strategy on how to study this issue and show the early results of my work.
